Study on the mechanism of metformin regulating osteogenic differentiation of bone marrow mesenchymal stem cells in diabetic rats by up-regulating BMP-2/IGF-1 expression
WANG Yang;GUO Li;KE Ruiqiong;LIU Dengke;WU Dan;Objective : To investigate the effects of different concentrations of metformin on osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs) in diabetic rats in vitro and its molecular mechanism. Methods : Diabetic male Wistar rat models were established by intraperitoneal injection of streptozotocin(STZ, 40 mg/kg). BMSCs were isolated, cultured, and the third-generation cells were used for the experiments. The cells were randomly divided into three groups: control group(without metformin), low-dose group(10 μmol/L metformin), and high-dose group(100 μmol/L metformin). All groups were cultured in osteogenic induction medium. The alkaline phosphatase(ALP) activity of the cells was detected on the 7 th and 14 th days of induction differentiation, and alizarin red staining was used to observe the formation of calcified nodules. The expression levels of osteogenic-related proteins bone morphogenetic protein-2(BMP-2) and insulin-like growth factor-1(IGF-1) were also detected by protein immunoblotting. Results : On the 7 th and 14 th days of induction differentiation, compared with the control group, the ALP activity and the formation of calcified nodules increased in the low-dose and high-dose metformin groups, and the high-dose group was superior to the low-dose group. Compared with the control group, both low-dose and high-dose groups showed increased protein expression levels of BMP-2 and IGF-1(P<0. 05), with the high-dose group exhibiting higher protein expression levels of BMP-2 and IGF-1 than the low-dose group(P<0. 05). Conclusion : Metformin can dose-dependably enhance the osteogenic differentiation ability of BMSCs in diabetic rats, and the mechanism may be related to the upregulation of BMP-2 and IGF-1 protein expressions.
Predictive value of serum parameters, fetal umbilical artery blood flow parameters, and growth indicators for pregnancy outcomes in pregnant women with gestational diabetes mellitus
CHEN Limin;CHEN Chu;YE Mei;YE Shengfu; Objective : To analyze the predictive value of serum indicators, fetal umbilical artery blood flow parameters and fetal growth indicators for pregnancy outcomes in pregnant women with gestational diabetes mellitus(GDM). Methods : A total of 300 pregnant women with GDM admitted to Guangdong Clifford Hospital from June 2022 to June 2025 were enrolled in this retrospective study. According to pregnancy outcomes, the subjects were divided into adverse outcome group and favorable outcome group. General data, serum indicators, fetal umbilical artery blood flow parameters and fetal growth indicators were collected and compared between the two groups. Logistic regression analysis was performed on the statistically different indicators to identify the independent risk factors for adverse pregnancy outcomes in GDM women. A nomogram and receiver operating characteristic(ROC) curve were drawn to evaluate the predictive value of these factors for adverse pregnancy outcomes in pregnant women with GDM. Results : Among the 300 GDM pregnant women, 46 cases had adverse pregnancy outcomes(adverse outcome group), and the remaining 254 cases had favorable pregnancy outcomes(favorable outcome group). The levels of fasting plasma glucose(FPG), fetal umbilical artery pulsatility index(PI), systolic/diastolic ratio(S/D), abdominal circumference(AC) and estimated fetal weight(EFW) in the adverse outcome group were significantly higher than those in the favorable outcome group(P<0. 05), and these indicators were independent risk factors for adverse pregnancy outcomes in GDM women. The nomogram combined with calibration curve showed that the predictive model established on the above factors had higher application value. The area under the curve(AUC) of the ROC curve of the combined prediction for adverse pregnancy outcomes was 0. 875(95% CI: 0. 821-0. 929). Conclusion : Fasting plasma glucose, PI, S/D, AC and EFW are independent risk factors for adverse pregnancy outcomes in pregnant women with GDM. The nomogram prediction model constructed based on the above indicators has higher predictive value, and combined detection of the five indicators can improve the predictive accuracy of adverse pregnancy outcomes in GDM pregnant women.
The role of polysaccharide from the seed cakes of Camellia oleifera Abel in inhibiting the development of murine colitis
XI Yuling;HE Liangmei;WANG Xiaoyin;LIU Zhiping; Objective : To investigate the preventive and therapeutic effects of polysaccharides from the seed cakes of Camellia oleifera Abel(COCP) on colitis in mice. Methods : C57 BL/6 mice were randomly divided into five groups: a negative control group, a colitis model group, and COCP intervention groups receiving low(25 mg/kg), medium(50 mg/kg) and high(100 mg/kg) doses. An experimental colitis model was induced using 3% sodium dextran sulphate(DSS); COCP was administered prophylactically for 14 consecutive days via gastric lavage, starting from 7 days prior to model induction and continuing until 7 days after induction; the disease activity index(DAI) scores were calculated, and changes in inflammatory cytokines, tight junction molecules(both gene expression and protein levels) in colonic tissue, and faecal microbiota were assessed; bone marrow-derived macrophages(BMDMs) were isolated from C57 BL/6 mice and cultured in vitro. The cells were divided into five groups: a negative control group, an inflammatory model group, and COCP intervention groups at low(3. 9 μg/mL), medium(7. 8 μg/mL) and high(15. 6 μg/mL) concentrations. An in vitro inflammatory model was established by stimulating BMDMs with lipopolysaccharide(LPS); the COCP intervention groups at each concentration were treated with the corresponding concentration of COCP, and the effects of COCP on the BMDM inflammatory model were observed; transcriptomic sequencing was used to analyse the molecular mechanisms of COCP. Results : Compared with the colitis model group, following COCP intervention, the rate of weight loss in mice was markedly slowed, the DAI was significantly reduced, colon length increased, and histopathological damage to the colon was alleviated. Furthermore, COCP downregulated the mRNA expression levels of inflammation-related cytokines(Il6, Tnf and Ccl2) in colonic tissue and inhibited the phosphorylation of NF-κB p65; In an LPS-induced BMDM inflammation model, compared with the inflammation control group, COCP suppressed the gene expression levels of Tnf and Ccl2. Mechanistically, COCP increased the number of goblet cells in colonic tissue and upregulated the gene and protein expression levels of tight junction molecules such as E-cadherin and Occludin; it also modulated the gut microbiota composition, increasing the abundance of beneficial bacteria such as Tannerellaceae and Akkermansiaceae. Transcriptomic sequencing revealed that COCP significantly upregulates the expression of genes such as Gm42031, Rps3a2 and Grm4, whilst downregulating the expression of genes such as Saa1, Gm21887 and Cyp2d26. Conclusion : COCP inhibits colitis in mice, probably exerting its inhibitory effect by suppressing the inflammation level, enhancing the barrier function, regulating the ecology of the flora, and improving the DNA repair ability.
Investigations on the role and mechanisms of Telaglenastat in pathological cardiac hypertrophy
WANG Kun;LIU Ruobing;LI Hongliang;HU Yufeng; Objective : To investigate the therapeutic effect of the small molecule compound Telaglenastat on pathological myocardial hypertrophy, aiming to provide a potential lead compound and research direction for the clinical development of related therapeutic agents. Methods : H9C2 and AC16 cells were treated with different concentrations of Telaglenastat, and the CCK-8 assay was used to evaluate the cytotoxic effects of telaglenastat on normal cardiomyocytes. A phenylephrine(PE)-induced neonatal rat cardiomyocyte hypertrophy model was established, and the cells were divided into four groups(DMSO+PBS, DMSO+PE, 2. 5 μmol/L Telaglenastat+PE, and 5 μmol/L Telaglenastat+PE) for efficacy evaluation. The mRNA and protein expression levels of the cardiac hypertrophy markers atrial natriuretic peptide(Anp) and B-type natriuretic peptide(Bnp) were detected by RT-qPCR and western blot, respectively. Transcriptomic analysis was performed to investigate the effects of telaglenastat on signaling pathways related to cardiac hypertrophy and its underlying mechanisms. A mouse model of transverse aortic constriction(TAC)-induced pathological cardiac hypertrophy was established, and the mice were divided into four groups: sham surgery + vehicle(Sham+Vehicle), sham surgery + Telaglenastat(Sham+Telaglenastat), TAC surgery + vehicle(TAC+Vehicle), and TAC surgery + Telaglenastat(TAC+Telaglenastat). Cardiac function in the four groups was assessed by echocardiography. Hematoxylin-eosin(HE) staining and picrosirius red(PSR) staining were used to observe histopathological changes in heart tissues from the four groups. Results : CCK-8 assay results showed that, compared with the DMSO control group, there was no significant cytotoxicity in normal cardiomyocytes( H9C2 and AC16) treated with various concentrations of Telaglenastat(P>0. 05), except that a statistically significant difference in cell viability was observed in AC16 cells treated with 5 μ mol/L Telaglenastat(P<0. 001). In the PE-induced cardiomyocyte hypertrophy model, compared with the DMSO+PE group, the Telaglenastat+PE group showed significantly downregulated mRNA and protein expression levels of the cardiac hypertrophy markers Anp and Bnp(P<0. 01). In the TAC mouse model, compared with the TAC+Vehicle group, the TAC+Telaglenastat group exhibited significantly increased left ventricular ejection fraction(EF) and fractional shortening(FS)(P<0. 05), as well as significantly reduced heart weight-to-body weight ratio(HW/BW), lung weight-to-body weight ratio(LW/BW), and heart weight-to-tibia length ratio(HW/TL)(P<0. 01). HE and PSR staining results showed that, compared with the TAC+Vehicle group, the TAC+Telaglenastat group had smaller cardiomyocyte cross-sectional areas and less cardiac collagen fibrosis. Transcriptomic analysis revealed that, compared with the DMSO+PE group, the Telaglenastat+PE group exhibited downregulated expression of genes related to inflammatory response, fibrosis, and protein synthesis, along with upregulated expression of genes involved in the mitophagy pathway. Conclusion : Telaglenastat effectively inhibits pathological cardiac hypertrophy and improves cardiac function both in vitro and in vivo, and its protective effects may be associated with the activation of the mitophagy pathway in cardiomyocytes.

XIANG Youfu;XIONG Shiyin;QIU Xiufeng;LU Puyang;MIN Lingfeng;WU Xinhao;YOU Cong; The First Clinical Medical School of Gannan Medical University; Objective : To explore the causal relationship between inflammatory factors and atopic dermatitis(AD), allergic rhinitis(AR), and allergic asthma(AA) using a two sample Mendelian randomization(TSMR) analysis method. Methods : Data was collected on 41 inflammatory factors from a genome-wide association study(GWAS) as exposure and data on AD, AR, and AA from the finnGen study as outcomes. The TSMR analysis, we employed the inverse-variance weighted(IVW) method, the weighted median(WM) method, and the MR-Egger regression method to infer causality. Several sensitivity analysis methods were used to assess the stability of the results, and the MR Steiger test was used to determine the correctness of the causal direction. Results : The TSMR analysis revealed that macrophage colony-stimulating factor(M-CSF) was associated with an increased risk of AD(IVW:OR=1. 053, 95%CI=1. 001-1. 107, P=0. 044), Interleukin-5 was associated with an increased risk of AR(IVW:OR=1. 078, 95%CI=1. 000-1. 161, P=0. 049), Regulated upon activation, Normal T cell expressed and secreted(RANTES) was associated with a reduced risk of AR(IVW:OR=0. 922, 95%CI=0. 856-0. 994, P=0. 034), and granulocyte colony-stimulating factor(G-CSF) was associated with a reduced risk of AA(IVW: OR=0. 880, 95%CI=0. 782-0. 990, P=0. 033). Sensitivity analyses confirmed the robustness of these results, and the MR Steiger test did not detect reverse causality. Conclusion : M-CSF may be an upstream risk factor for atopic dermatitis, IL-5 may be an upstream risk factor for allergic rhinitis, RANTES may be an upstream protective factor for allergic rhinitis, and G-CSF may be upstream protective factors for allergic asthma.
Experimental Study on Anti-inflammatory effect of Osthol
Lian Qishen; Zhou Li; Ye Heyang; et alOsthol(Ost) was a coumarin isolated from the fruits of Cnidium monnieri(L ) Cusson, Ostsuppressed the swelling of mouse ears induced by zylene and the increase of peritoneal caPillarypermeability by ip 0.7% acetic acid in mice. Ost diminished the ederna of hind paws induced by sc1% carrageenan not only in normal rats, but also in adrenalectomised rats. However, the conentsof PGE in the inflammtory tissue of rat hind paws was not changed. Furthermore, Ost inhibitedthe proiferation of granulotna induced by impanting 0.1 ml agar in mice. These results suggestedthat Ost had anti-inflammatory effects, which was neither dependent on the pituitary-adrenalsystem nor on the PGE synthesis.

Current situation and progress of rehabilitation treatment of autism
QU Jia-yang;HE Zheng-yi;LI Lin-cai;ZOU Zheng-wei;WANG Mao-yuan;YE Jun-son;Gannan Medical University;Autism spectrum disorder is a neurodevelopmental disorder characterized by repetitive behaviors, social dysfunction, and interest stenosis. The pathogenesis of autism has not been clear up to now,and there is no specific medicine. The increasing number of people with autism in China has brought great pressure to the family and society.Appropriate treatment will improve the core symptoms of autistic children, and it is very important to choose the appropriate method among many intervention means. However,at present, there is chaos and problems in the autism rehabilitation field, and the professionalism of various institutions in the rehabilitation field is uneven. This paper will discuss and summarize several intervention methods and their effects. At present,in addition to the conventional treatment methods utilized by rehabilitation institutions, speech therapy, sentiment training, behavior intervention and structured training, art therapy, peer support therapy and animal-assisted therapy are also gradually emerging, making up for the shortcomings of traditional training methods. As a new approach to treat autism based on cell technology, stem cell therapy for autism is one of the hot spots in current autism treatment research, which is expected to effectively improve the core symptoms of autism. Given the heterogeneity of autism,a single treatment is not desirable. The injection therapy of stem cells and the intervention of various rehabilitation treatment methods will be a new model of autism treatment in the future.
